AIMS: The ReQuest database is the basis for the investigation of different aspects of GERD symptomatology and the treatment induced symptom changes. By using the dimension acid complaints which influences the patient's general well-being highly, treatment effects of PANTO and OME were investigated.
METHODS: The ReQuest database includes data from all clinical trials with ReQuest, ie, 14 trials with 8,177 (6,810) patients in the intention to treat (per protocol; PP) population. For this analysis, all patients treated with PANTO 40 mg once daily (od) or OME 20 mg od for 28 days who filled in the ReQuest daily were chosen. As in the OME group only patients with GERD grade B to D (LA classification) were available, the PANTO data set was restricted to comparable patients. For both treatment groups the median number of acid episodes during the 28 days of treatment was evaluated. Data sets were analyzed for all patients as well as separated for the different GERD grades. Comparisons between the treatment groups were done by Wilcoxon two-sample test.
RESULTS: For this analysis, 457 patients treated with OME 20 mg od and 1041 patients treated with PANTO 40 mg od were eligible (PP). Overall, during the 28 days of treatment, patients in the PANTO (OME) group experienced 21.5 (33.0) acid episodes; difference proved to be statistically significant. For GERD grade B and C significantly less acid episodes were observed in the PANTO group. For GERD grade D, patient numbers were too low to permit statistical meaningful analyses. In-group comparison between the GERD grades revealed only minor differences.

CONCLUSIONS: During 28 days of treatment patients treated with panto 40 mg OD experienced signficantly less acid episodes than patients treated with OME 20 mg. The in-group comparison showed similar numbers of acid episodes for the difference GERD grades.
